... use the DS2416 card's software to route the audio from the second SW1000XG card to the first one...

That doesn't make sense, I'm afraid... The software only controls the DS2416's onboard DSP and the signal routing between its own inputs, auxes, busses and outputs. It also controls the connections between the DS2416 wave drivers and the input channels and busses. It is completely separate from the SW1000XG wave drivers, however, and can't magically "route" audio back to it in the way you seem to describe. Please note that the internal ribbon cable is a one-way connection (SW1000XG output to DS2416 input).

Yes, it is possible to connect the DS2416 analog outputs to the SW1000XG's mic/line input, and route audio back that way. This mostly serves to complicate the signal flow, though; The same cable could instead be used to connect the two SWs directly, and this would in fact be a preferred setup if the DS2416's processing power isn't needed.

If you already have a DS2416 and the software to control it, and also want to make good use of it, I would argue that connecting both SW cards (one via analog I/O, one via SPDIF or internal ribbon) to the DS inputs is a more elegant solution. This way, once the DS control software is set up properly, audio from the SW cards appear on separate input channel strips (along with other available sources, such as wave data from a DAW), and can be mixed and processed freely. Once the mix is ready, it can be recorded via the DS busses/wave drivers. I would recommend connecting the speakers/monitoring system to the DS outputs, since it is now the endpoint of the audio chain.
  • The default DS2416 configuration has input sources for channels 1-16 set as "PCI PB" (audio playback from software via the drivers), channels 17-18 for the rear analog inputs, channels 19-20 for the S/PDIF input and channels 21-24 as returns from the two onboard stereo effects.
  • To use the rear S/PDIF input, the DS2416 wordclock must be set to "S/PDIF".
  • To use the internal ribbon cable from a SW1000XGs, the DS2416 wordclock must be set to "SERIAL" or "SUB" (depending on the software). You must also switch the input source on up to 8 mixing channels to "SI" (Serial or Sub Input).
  • The DS2416 is equipped with 4 pairs of "PCI REC" outputs (audio recording sources for your software via the drivers). In the default configuration, the Main L/R bus is routed to all PCI REC pairs, which means that you can record a stereo mixdown without changes to the routing.
